目的探讨氨溴索对表皮葡萄球菌(S.epidermidis)物被膜(biofilm,BF)主要成分胞间多糖黏附素(polysaccharideintercellularadhesion,PIA)的干预作用,研究其对PIA合成过程中重要基因表达的影响。方法平板法培养表皮葡萄球菌RP62A24h,得到成熟BF,不同浓度氨溴索作用24h后,将BF振荡下来,用硫酸一苯酚法检测氨溴索对PIA含量的影响;RT-PCR检测氨溴索对PIA合成过程中起重要作用的基因icaA、icaR、泸和,龇s的表达的影响。结果1.875mg/mL氨溴索作用组和对照组的PIA含量(~g/10mg)分别为(345.1050±8.9603)和(403.6269±16.9947),t=-7.213,P〈0.05;icaAmRNA的表达(A/对照)分别为(0.4084±0.0461)SH(0.5540±0.0341),t=6.008,P〈O.05;icaRmRNA的表达(A/对照)分别为(0.4253±0.0513)和(0.2826±0.0636),t=13.066,P〈0.05;mRNA的表达(A/对照)分别为(0.4236±0.0258)和(0.5291±0.0246),t=-6.968,P〈0.05;luxSmRNA的表达(A/对照)分别为(0.2947±0.0367)和(0.1783±0.0385),t=5.180,P〈0.05。3.75mg/mL氨溴索作用下有同样的趋势且效应更加明显。结论氨溴索可降低表皮葡萄球菌BFPIA的含量和其调控基因icaA、和泸的mRNA的表达,增加fc积和,姒s的mRNA的表达。
Objective To explore the effect of ambroxol on polysaccharide intercellular adhesion(PIA) of Staphylococcus epidermidis biofilm(BF) in an established biofilm model in vitro. Further to investigate the effect ambroxol on the expression of important genes for synthesis of PIA. Methods The strain RP62A of Staphylococcus epidermidis was incubated for 24h to form mature BF on slide. After treated with ambroxol of different concentrations for 24h, the BF was oscillated and collected for investigation. The effect of ambroxol on PIA was detected through the sulfuric acid-phenol method. The mRNA expression of genes which are important to the synthesis of PIA such as icaA, icaR, ~ and luxS were measured by reverse transcription PCR. Results PIA content(gg/10mg) of the 1.875mg/ mL ambroxol treatment group and control group were (345.1050±8.9603) and (403.6269±16.9943), respectively,t=-7.213,P〈0.05; The mRNA expression of icaA (A/control) were (0.4084±0.0461) and (0.5540±0.0341) ,respectively, t=6.008, P〈0.05: The mRNA expression of icaR (A/control)were (0.4253±0.0513)and (0.2826±0.0636), respectively, t=-I 3.066, P〈0.05: The mRNA expression of M (A/control) were (0.4236±0.0258) and (0.5291±0.0246), respectively, t=6.968, P〈0.05: The mRNA expression of luxS (A/control)were (0.2947±0.0367) and (0.1783±0.0385), respectively, t=5.180, P〈0.05.The effect of ambroxol with the concentration of 3.75mg/mL had the same impact but more obvious. Conclusion Ambroxol can decrease the content of PIA and the mRNA expression of icaA and OB and increase the mRNA expression of icaR and luxS, which effect was positively relevant to the concentration of ambroxol.
