柑橘和黄皮中黄龙病菌含量动态变化

【目的】对柑橘和黄皮样品中黄龙病(Huanglongbing)病原菌含量进行实时定量检测,明确其病原菌分布及动态变化规律,为黄龙病的综合防控提供参考依据。【方法】分别采用常规PCR和STDP PCR检测22个柑橘、黄皮和九里香样品中的黄龙病菌。在不同时期采集夏橙、沙田柚和黄皮的叶脉、枝、根、果实中柱(黄皮取果蒂)样品,采用STDP PCR进行实时定量检测。【结果】黄皮、九里香样品采用常规PCR均未检测出黄龙病菌,通过STDP PCR检测各有2个样品为阳性;柑橘样品采用常规PCR检测有2个样品呈阳性,通过STDP PCR检测有4个样品为阳性。在黄皮中不同时期黄龙病病菌含量均较低,且无明显变化规律,叶脉和枝条上黄龙病菌检出率明显高于根部,在果蒂部分未检出黄龙病菌;夏橙和沙田柚中根部黄龙病菌含量明显低于叶脉、枝及果实中柱,且在9-12月黄龙病菌含量高于其他时期。【结论】柑橘、黄皮和九里香均是柑橘黄龙病的寄主,生产上可参考柑橘在不同时期的黄龙病菌含量变化确定防控方式和时期。

英文摘要:

【Objective】In order to provide references for synthetically preventing Huanglongbing, the present experiment was conducted to detect Huanglongbing pathogen content in citrus and wampee by real-time quantitative detection to investigate distribution and dynamic variation of Huanglongbing pathogen. 【Method 】The Huanglongbing pathogen in 22 samples of citrus, wampee(Clausena lansium) and jasmine orange(Murraya paniculata) was detected by conventional PCR and STDP PCR techniques. The leaf vein, branch, root, fruit central cylinder(pedicle of wampee) samples were detected by STDP PCR technique, which were collected from Valencia orange(Citrus sinensis Osbeck), Shatian pomelo(Citrus grandis Osbeck) and wampee at different periods. 【Result】The results showed that Huanglongbing pathogen wasn't detected out in all wampee and jasmine orange samples by conventional PCR technique, however, 2 samples each were identified to be positive by STDP PCR technique. In addition, among all citrus samples, 2 samples were identified to be positive by conventional PCR technique, while 4 samples were identified to be positive by STDP PCR technique. Furthermore, the content of pathogen in wampee was relatively lower at different period without obvious change law, while the positive rates of pathogen in leaf vein and branch were significantly higher than that in root, and the pathogen wasn't detected out in pedicle of wampee. As for Valencia orange and Shatian pomelo, the content of pathogen was significantly lower in root than that in leaf vein, branch and central cylinder, and significantly higher from September to December than that at other periods. 【Conclusion 】Citrus, wampee and jasmin orange are host plants of Huanglongbing pathogen.And the changes of pathogen content in citrus can provide references for prevention method and period of Huanglongbing.
