目的探讨天然牛磺酸联合中药对肝星状细胞的影响。方法体外培养大鼠肝星形细胞(HSC-T6),将其分成空白对照组、天然牛磺酸组、中药组、天然牛磺酸联合中药组、秋水仙碱组,按各组要求往细胞孔内加入相应的含药血清,培养孵育48 h后,用四氮噻唑蓝(MTT)比色法测定细胞的吸光度值,用流式细胞仪检测各周期细胞的DNA含量及细胞凋亡情况。结果秋水仙碱组、天然牛磺酸组、中药组、天然牛磺酸联合中药组HSC-T6细胞光密度值(OD)均显著低于空白对照(P〈0.05);天然牛磺酸联合中药组对HSC-T6细胞增殖抑制率明显高于天然牛磺酸组和中药组(P〈0.05);天然牛磺酸组和中药组对HSC-T6细胞增殖抑制率与秋水仙碱组比较差异均无统计学意义(P〉0.05);天然牛磺酸联合中药组与秋水仙碱组对HSC-T6细胞增殖抑制率比较差异有统计学意义(P〈0.01);秋水仙碱组、天然牛磺酸组、中药组、天然牛磺酸联合中药组HSC-T6细胞G0/G1期DNA百分比均显著高于空白对照组(P〈0.05);天然牛磺酸组、中药组、天然牛磺酸联合中药组和秋水仙碱组HSC-T6细胞S期、G2/M期DNA百分比均显著低于空白对照组(P〈0.05);天然牛磺酸联合中药组HSC-T6细胞G0/G1期DNA百分比高于天然牛磺酸组和中药组(P〈0.05);天然牛磺酸联合中药组HSC-T6细胞S期、G2/M期百分比均低于天然牛磺酸组和中药组(P〈0.05);天然牛磺酸联合中药组HSC-T6细胞G0/G1期DNA百分比高于秋水仙碱组(P〈0.05);天然牛磺酸联合中药组HSC-T6细胞S期和G2/M期DNA百分比均低于秋水仙碱组(P〈0.05);空白对照组HSC-T6细胞晚期凋亡率为(7.346±0.836)%,低于秋水仙碱组、天然牛磺酸组、中药组、天然牛磺酸联合中药组的(19.647±1.009)%、(24.178±0.996)%、(23.882±1.105)%和(31.533±1.002)%(P〈0.05);天然牛磺酸联合中药组HSC-T6细胞晚期凋?
【Objective】To investigate the effects of natural taurine combined with Chinese medicine on hepatic stellate cells(HSC).【Methods】HSC-T6 cell lines of rats cultivated in vitro were divided into blank control group,natural taurine group,Chinese medicine group,natural taurine combined with Chinese medicine group and colchicine positive control group,and the serum containing medicine was added to each cell hole according to the requirements of each group.After 48 hours of cultivation,the cell absorbance value was measured by four nitrogen thiazole blue colorimetric method,and flow cytometry instrument was used to detect the DNA content and cell cycle,cell apoptosis.【Results】Compared with the blank control group,the optical density(OD) value of HSC-T6 cell in colchicine positive control group,natural taurine group,Chinese medicine group and natural taurine combined with Chinese medicine group decreased significantly(P〈0.05);the inhibition of proliferation of HSC-T6 cells in natural taurine combined with Chinese medicine group was significantly higher than that in natural taurine group and Chinese medicine group(P〉0.05);there was no significant difference in the inhibition of proliferation of HSC-T6 cells between natural taurine group,Chinese medicine group and colchicine positive control group(P〈0.05);there was a significant difference in the inhibition of proliferation of HSC-T6 cells between natural taurine combined with Chinese medicine group and colchicine positive control group(P〈0.01);compared with the blank control group,the percentage of DNA in HSC-T6 cells in G0/G1 phase of the colchicine positive control group,natural taurine group,Chinese medicine group,natural taurine combined with Chinese medicine group was significantly increased(P〈0.05);the percentag of DNA in HSC-T6 cells in S phase,G2/M phase of natural taurine group,Chinese medicine group,natural taurine combined with Chinese medicine group and colchicine positive control group was significantly lower
