目的利用基因芯片技术研究“益气调血,扶本培元”针法对快速老化小鼠SAMP10皮质基因表达的影响。方法选取雄性8月龄快速老化小鼠SAMP10(分成针刺腧穴组,对照组)和同源正常老化小鼠SAMR1,采用cDNA microarray技术,分析伴随快速老化SAMP10皮质基因表达谱的变化以及针刺对其的影响。结果芯片结果显示,伴快速老化。P10皮质共有9类17个基因的表达发生变化,15个基因表达下调,2个基因表达上调,施予“益气调血,扶本培元”针法后,所有与P10快速老化相关基因的表达均被部分或全部的逆转,显示针刺具有特异性。结论“益气调血,扶本培元”针法能够良性调节脑老化相关基因的表达,并可能通过该种作用延缓脑衰老。
Objective To investigate the influence of acupuncture method of “Replenishing qi to regulate blood and reinforcing the essence to strengthen the body” on cortical gene expression by a gene chip technique in swift aging mice (SAMP10). Methods Male 8-month-old swift aging mice (SAMP10) and isogenous normal aging mice (SAMR1) were selected to observe changes with swift aging in SAMP10 cortical gene expression spectrum by a cDNA microarray technique and to investigate the influence of acupuncture on them. Results The chip results showed that with swift aging, the expressions of 17 genes of 9 kinds changed in P10 cortex, with 15 genes down-modulated and 2 genes up-modulated. After acupuncture method of “Replenishing qi to regulate blood and reinforcing the essence to strengthen the body” was used, all P10 swift aging-related gene expressions were reversed partly or totally, showing that acupuncture had its specificity. Conclusion Acupuncture method of “Replenishing qi to regulate blood and reinforcing the essence to strengthen the body” can beneficially modulate the expression of cerebral aging-related gene and may thereby delay cerebral senility.
