目的:探讨CD98与炎症性肠病( IBD)的关系。方法:雄性Balb/c小鼠48只,分为4组。正常对照组:不进行干预,自由饮食。生理盐水组:灌肠前禁食24 h,自由饮水,于实验第1、8及15天分别用100μL的生理盐水灌肠,灌肠后自由饮食。乙醇组:用体积分数50%乙醇溶液100μL灌肠,方法和步骤同生理盐水组。 TNBS+乙醇组:第1、8及15天分别用2.0、2.5和3.0 mg TNBS加体积分数50%乙醇溶液100μL灌肠,建立TNBS诱发的小鼠实验性IBD模型。实验第21天,采用流式细胞仪检测外周血CD98的表达,免疫组化SABC法检测结肠黏膜上皮组织中CD98蛋白的表达。结果:4组小鼠外周血和结肠黏膜上皮组织中CD98蛋白表达的差异均有统计学意义(F=29.460,χ2=4.567,P<0.05),TNBS+乙醇组高于其他3组。结论:CD98可能参与了IBD的发生发展。
Aim:To explore the pathogenic role of CD98 in inflammatory bowel disease(IBD).Methods:The IBD model in mice was induced by TNBS .Forty-eight male Balb/c mice were randomly divided into four groups (n=12).Nor-mal control group:mice were given an unrestricted diet .Normal saline group: mice were put in a fasted state for twenty-four hours,but could drink water free , then given unrestricted diet after having received a 100 μL normal saline enema on the first,eighth,and fifteenth day of the experiment .Ethanol group:mice were given 100 μL 50%solution ethanol enema , other steps were the same to the normal saline group .TNBS+ethanol group:mice were respectively instilled by 2.0,2.5, and 3.0 mg TNBS dissolved in 100 μL 50%ethanol enema on the first , eighth,and fifteenth day of the experiment .The CD98 in peripheral blood and colonic epithelial tissue were detected by flow cytometry and immunohistochemistry ,respec-tively.Results:The expression of CD98 in peripheral blood and colonic epithelial tissue significantly increased in the TN-BS+ethanol group as compared with the other three groups (F=29.460,χ2 =4.567,P〈0.05).Conclusion:CD98 may be involved in the pathogenesis of IBD .
