目的:观察左归丸、右归丸对体外培养的成骨细胞骨硬化蛋白(Sclerostin)及β连环蛋白(β-catenin)表达的影响。方法:成年SD大鼠灌胃法获得左归丸、右归丸含药血清。体外培养SD乳鼠颅骨成骨细胞,取第3代细胞分对照组、左归丸组、右归丸组,各组含药血清培养后分别测细胞增殖、Sclerostin和β-catenin蛋白表达。结果:与对照组比较,右归丸组在5d、7d、9d时细胞明显增殖,左归丸组在7d、9d时细胞显著增殖(P〈0.01),与左归丸组比较,右归丸组在5d时细胞明显增殖(P〈0.05)。右归丸组在24h、48hSclerostin蛋白表达较对照组增高(P〈0.05),左归丸组在24h较对照组减少(P〈0.05)。与右归丸组相比,左归丸组在24h、48h Sclerostin蛋白表达显著下降(P〈0.01)。右归丸组β-catenin蛋白表达在48h时较对照组有显著增高(P〈0.05),左归丸组β-catenin蛋白表达在24h、48h均显著高于对照组(P〈0.01)和右归丸组(P〈0.05)。结论:右归丸、左归丸对成骨细胞均具有显著的促增殖作用,而右归丸较左归丸作用更明显。左归丸可能是通过抑制Sclerostin蛋白的表达发挥促成骨作用,右归丸无抑制Sclerostin蛋白表达作用。
Objective: To investigate the effects of medicated serum of Yougui Pill and Zuogui Pill on the proliferation and expression of Sclerostin and 13-catenin of osteoblast. Methods: Osteoblast were obtained from the new born SD rats and cultured in vitro. The passage three cells were used. They were divided into 3 groups: control group, Yougui Pill group and Zuogui Pill group. To observe the influence on the proliferation and the expression of Sclerostin and ~-catenin of osteoblast after incubated. Results: The proliferation of osteoblast was increased in each experiment group, compared with the control groups, there were significant differences in Yougui Pill group at 5, 7 and 9 days, and in Zuogui Pill groups at 7 and 9 days (P〈0.01). Compared with the control group, the expression of Sclerostin of osteoblast were increased in Yougui Pill group at 24h and 48h (P〈0.05), and decreased in Zuogui Pill group at 24h and 48h. There were significant differences at 24h (P〈0.05) in Zuogui Pill group, also had significant differences compared with Yougui Pill group(P〈0.01). The expression of [I-catenin of osteoblast were all increased in both Yougui Pill groups and Zuogui Pill groups at 24h and 48h. There were significant differences compared with Yougui Pill group (P〈0.05). Conclusion: Yougui Pill and Zuogui Pill can significantly increase the proliferation of osteoblast, Yougui Pill was better than Zuogui Pill. And only Zuogui Pill can significantly decrease the expression of Sclerostin of osteoblast. This might be the mechanism of promoting bone formation of Zuogui Pill.
