目的:建立苯酚-硫酸法测定云南部分兰科植物中的多糖含量。方法:用超声法提取石斛多糖,用苯酚-硫酸法进行含量测定,以葡萄糖为对照品,检测波长为487 nm,用L_9(3~4)正交试验设计优化测定条件。结果:加5%苯酚液1.0 m L、硫酸5.5 m L,40℃水浴显色25 min,测得叠鞘石斛中多糖含量为11.896 mg/g。结论:优化后的苯酚-硫酸法测定兰科植物中多糖含量方法简单,重复性好,结果准确。
Objective: Determination of Orchidaceae plants polysaccharide content established by phenol sulfuric acid method. Method of ultrasonic extraction was used to extract polysaccharides from Ulva, were determined by phenol sulfuric acid method, using glucose as the standard, detection wavelength was 487 nm, L9 (34) orthogonal design to optimize the determination condition. The sulfuric acid and phenol solution 1.0 mL of 5 % and 5.5 mL, 40 DEG C water bath heat preservation 25 min results best measured Ulva ultrasonic extract polysaccharide content was 11.896 mg/g Conclusion the optimized phenol - sulfuric acid method for determination of Orchidaceae plants polysaccharide content is simple, reproducible, accurate.
