ObjectiveTo optimize the extraction technology of total amino acids from parts of Orchidaceae plants in Yunnan anddetermine the content. Methods：The best extraction conditions were optimized by orthogonal test,and the content of total aminoacids was determined in 568 nm through Nihydrin colorimetry method. Results：The best extraction conditions were as follows： 20 mL75% ethanol,3 times of reflux extraction and 60 min for each time. The concentration and the absorbance of the GLY show a goodlinear correlation when changing from 0.5 μg/mL to 10 μg/mL,r = 0.999 8. Conclusion：Nihydrin colorimetry method is simple,accurate,which can be used for the determination of total amino acids in Orchidaceae plant.
