目的观察灯盏细辛注射液对损伤心脏微血管内皮细胞(CMEC)的保护作用及其对合成释放炎性介质的影响,寻找灯盏细辛注射液的作用靶点和机制,为临床应用提供实验依据。方法本实验采用大鼠乳鼠CMEC为研究对象,肿瘤坏死因子(TNF-α)为刺激剂,检测CMEC细胞活力(MTT)及乳酸脱氢酶(LDH)释放量;用双抗体夹心酶联免疫法(ELISA)检测损伤的CMEC释放白介素6(IL-6)、单核细胞趋化蛋白1(MCP-1)的含量;用实时定量PCR方法检测损伤CMECIL-6、MCP-1mRNA的表达。结果灯盏细辛注射液(12.5,25,50和100mg·L^-1)提高TNF-α损伤CMEC的细胞活力,降低LDH的释放,灯盏细辛注射液(50和100mg·L^-1)抑制TNF-α损伤CMECIL-6和MCP-1的释放及mRNA的表达。结论灯盏细辛注射液能够抑制TNF-α诱导CMEC分泌IL-6、MCP-1的作用,降低其mRNA表达,减轻炎性因子对CMEC的损伤。灯盏细辛注射液减轻TNF-α对CMEC的损伤,抑制炎性因子的释放可能是其保护血管内皮细胞的作用机制之一。
OBJECTIVE To investigate the protective effects and mechanisms of Dengzhan Xixin injection on cardiac microvascular endothelial cells (CMECs) induced by TNF-α. METHODS After incubating CMECs with TNF-α (500 u?mL-1),cell vitality and cellular impairment were determined by MTT assay and lactate dehydrogenase (LDH) release. IL-6 and MCP-1 secretion were examined by ELISA. RT-PCR was employed to assess the mRNA expression of IL-6 and MCP-1. RESULTS Dengzhan Xixin injection groups (12.5,25,50 and 100 mg·L^-1 enhanced the vitality of CMECs and significantly inhibited the release of LDH. Dengzhan Xixin injection groups (50 and 100 mg·L^-1 reduced the secretion and mRNA expression of IL-6 and MCP-1 after 6 h incubation of TNF-α with CMEC. CONCLUSION Dengzhan Xixin injection could inhibit TNF-α-stimulated IL-6 and MCP-1 release and down-regulate mRNA expression of IL-6 and MCP-1 in CMECs. Dengzhan Xixin injection can attenuate damage of endothelial cells and may play an important role in suppressing flammation reaction induced TNF-α in CMECs.
