目的:研究川芎嗪(TMP)对不同剂量中波紫外线(UVB)诱导正常人黑素细胞黑素合成的影响,并初步探讨其机理。方法:常规分离培养正常人黑素细胞,分别用30mJ/cm^2、90mJ/cm^2的UVB照射细胞,然后加入100μg/ml、300μg/ml、600μg/ml的TMP孵育至72h,采用噻唑蓝(MTT)法测定细胞增殖情况;多巴氧化法测定酪氨酸酶活性;NaOH裂解法测定黑素含量。结果:30mJ/cm^2的UVB能诱导黑素细胞增殖,加强酪氨酸酶活性,促进黑素生成,TMP能显著抑制该效应,并呈剂量依赖性;90mJ/cm^2的UVB对黑素细胞产生明显的细胞毒作用,细胞增殖下降,黑素合成减少,TMP则能减轻其细胞毒作用,并也表现为剂量依赖性,以600μg/ml的TMP作用最为显著,差异有统计学意义。结论:TMP不但能抑制低剂量UVB照射诱导的黑素细胞增殖和黑素合成,而且还能减轻高剂量UVB照射对黑素细胞的细胞毒作用,对黑素细胞起一定的光保护作用。
Objective To investigate the effect of TMP on melanogenesis of melanocytes induced by different dosages of UVB. Methods A routine culture system of melanocytes was established, cells were irradiated with different dosages of UVB(30 and 90mJ/cm^2 respectively), and then incubated with different concentrations of TMP for 72h. MTT method, NaOH assay and DOPA oxidization reaction were employed to determine the cell proliferation, melanin synthesis, tyrosinase activity of melanocytes. Results 30mJ/cm^2 UVB could induce cell the proliferation, melanin synthesis, tyrosinase activity of melanocytes, TMP could inhibit this effect dose-dependently; 90mJ/cm^2 UVB showed toxicity to melanocytes, as the cell proliferation, melanin synthesis, tyrosinase activity of melanocytes were all downregulated, and TMP could retrieve this effect also in a dose-dependent manner. Conclusion TMP can not only inhibit the promoting effect on melanocytes induced by low-dosage UVB irradiation, but also retrieves the toxic effect induced by high-dosage UVB irradiation.
