目的:研究表没食子儿茶素没食子酸酯(EGCG)对中波紫外线(UVB)照射诱导永生化角质形成细胞株-Ha(W细胞的p53mRNA和p53蛋白表达的影响一方法:以一定剂量UVB照射HacaT细胞,并以200tc/mLEGCG处理照射后的HaCaT细胞,分别用RT-PCR法和Wester nblot方法检测各处理条件下p53mRNA和/或p53蛋白的表达水平。结果:30nd/cm^2的UVB照射后HacaT细胞的p53mRNA和p53蛋白表达逐渐增加,4h达到峰值,4h后随照射剂量增加而增加,24h后有所恢复;加入EGCG可下调UVB诱导的表达作用。结论:UVB照射对HaCaT细胞p53mRNA和p53蛋白的诱导表达有时效性与量效性,EGCG可下凋UVB照射的这种诱导作用:
Objective: To investigate the down regulation effects of epigallocatechin gallate (EGCG) on UVB induced p53 mRNA gene and protein expressions in HaCaT keratinocytes. Methods: HaCaT cells were cultured and irradiated with certain dosage of UVB, and then treated with 200 μg/mL of EGCG. RT - PCR and Western blot were used to detect p53 mRNA and p53 protein expressions. Results: The expressions of both p53 mRNA and p53 protein in HaCaT cells increased gradually and reached a peak at hour 4 and persisted almost for 24 h after the UVB irradiation of 30 mJ/cm^2 in a dose - dependence pattern ( P 〈 0,05 ). Alter addition of EGCG, the expression of p53 gene and p53 protein was down regulated ( P 〈 0,05 ), Conclusion: p53 ndtNA and p53 protein expressions of HaCaT cells are time- and dose- dependent on UVB inadiation, and EGCG can down regulate the p53 expression induced by UVB ircadiation.
