目的建立基于表面等离子体共振(SPR)生物传感器的解脲脲支原体(Uu)快速检测方法。方法将Uu的特异分子探针固定于传感器表面,应用SPR生物传感器实时在线分析系统对菌液中靶序列进行杂交检测,同时对该检测方法的灵敏度及特异性指标进行评价。结果当探针固定浓度为2.0μmol/L时,表面等离子体共振生物传感器检测Uu具有良好的检测效率,其检测灵敏度达10 fM,且检测其余3种阴性对照菌均无响应。结论 SPR传感器应用于Uu的快速检测具有敏感性高、特异性强等特点,具有良好的检测性能,检测方法准确可靠。
OBJECTIVE To establish a new Ureaplasma urealyticum detection technique based on surface plasmon resonance(SPR).METHODS The probe of U.urealyticum was immobilized on the SPR sensor surface.And the portable on line biomolecules analyzer based on SPR biosensor was applied to detect of the target sequences in the solution.Then the sensitivity and specificity of this method were investigated.RESULTS When fixed by probes at concentration of 2.0 μmol/L,it showed that the Ureaplasma urealyticum detection method had good sensitivity(10 fM) and specificity.The clinical test results compared with the conventional identification results were consistent.CONCLUSION The established U.urealyticum detection method has the advantages of high sensitivity,good specificity and good detection performance,which can be considered as an accurate and reliable detection method applied in the field of microbial detection.
