目的采用细胞悬液种植法和组织块移植法建立裸大鼠人乳腺癌皮下移植瘤模型并观察其生物学特性。方法体外培养人乳腺癌细胞株MDA-MB-231,注射于10只同种雌性裸大鼠皮下,2周后处死2只成瘤裸大鼠,将皮下瘤修剪成2mm。的组织块移植入10只同种雌性裸大鼠皮下。4周后将所有裸大鼠处死,取出皮下瘤。通过巨检、镜检、免疫组化等观察肿瘤的生物学特性。结果采用细胞悬液种植法的10只雌性裸大鼠皮下均成瘤,采用组织块移植法的10只雌性裸大鼠中8只皮下成瘤。两种方法的成瘤速度基本相似。HE染色和免疫组化染色证实两种方法建立的裸大鼠人乳腺癌模型的特性基本一致。结论采用细胞悬液种植法和组织块移植法均能成功建立裸大鼠人乳腺癌模型,为乳腺癌的体内实验和新型治疗等研究提供了较理想的动物模型。
Objective methods (injection of features. Methods To establish the subcutaneous human breast carcinoma models by two different cell suspensions and transplantation of tissues) and to observe their biological The subcutaneous tumor models were prepared by injection of cultured breast cancer cells suspensions directly or by transplantation of tumor tissue mass. MDA-MA-231 cell line was cultured in-vitro and inoculated subcutaneously into ten female nude rats. In 2 weeks 2 nude rates were sacrificed and the subcutaneous tumors were excised. The tumours were cut into small mass with volume of 2 mm3 , and then transplanted subcutaneously into another ten female nude rats. All nude rates were sacrificed in 4 weeks. The carcinoma mass were excised and studied through histopathology and immunohistochemistry. Results The subcutaneous tumors of the human breast carcinoma were generated in all 10 nude rats by injection of breast cancer cells suspensions,and in 8 of 10 nude rates by transplantation of tumor mass. The biological features of those 2 tumour models analyzed by HE staining and immunohistochemistry were showed with similarity as human breast carcinoma. Conclusions Both subcutaneous tumor models by the two different methods using nude rat can be served as ideal disease models in vivo study of human breast carcinoma.
