本研究采用大孔树脂混合使用技术,对桑叶总酚的富集工艺进行了优化。以总酚指纹图谱为指标,结合LC/MS技术和UV法,从8种不同型号的大孔树脂中优选出HPIM00和HPD826两种型号。对优选出的两种树脂的混合比例进行考察,并通过正交实验研究。最终确定这两种树脂的最佳混合比例为1:1(v/v),生药量与树脂比为0.8:1(g/mL)、上样浓度150mg/mL、上样流速10min/mL、60%乙醇洗脱,试验重复性良好。该技术可以较好保留总酚中各有效成分,保证药物疗效。优化出的工艺桑叶总酚得率高、简便易操作,适于工业的放大生产。
The enrichment process of total phenols from Folium Moil was optimized by mixed maeroporous resin. Based on the fingerprints of total phenols, the typical chromatograms recognized by LC/MS and the mass fraction of total phe- nols measured by UV, HPD400 and HPD826 were sifted out from eight types of maeroporous resin. The ratio of HPD400 and HPD826 was studied, and the optimal purifieation method was developed using orthogonal experiment. The optimized process was as follows:the ratio of HPD400 and HPD826 was 1:1 (v/v) ,the ratio of crude drug and macropourous resin was 0.8:1 (g/mL), the sample concentration was 150 mg/mL, the flow rate was 10 min/mL, the mobile phase was 60% ethanol. The results showed that the optimized process was simple, efficient and it provided good repeatability. This study provided a new approach for the separation and purification of total phenols from Folium Moil. It can be applied for large scale production in industry.
