目的:研究镰形棘豆总生物碱的体内抗肿瘤作用并探讨其作用机制。方法:采用S180荷瘤小鼠模型,将生理盐水和总生物碱灌胃给药,阳性药环磷酰胺腹腔注射,给药后检测小鼠的体质量、抑瘤率、胸腺指数以及脾指数,并通过HE染色,光镜下观察肿瘤细胞的形态变化。结果:镰形棘豆总生物碱高、中、低剂量组对S180荷瘤小鼠的抑瘤率分别为74.8%、79.3%、69.4%,总生物碱各剂量组对S180荷瘤小鼠的胸腺指数和脾指数均高于环磷酰胺组和生理盐水组,但无显著性差异。实验过程中,小鼠体质量没有降低,未显示任何毒性反应。HE染色发现,给药组肿瘤间质可见炎细胞浸润。结论:镰形棘豆总生物碱具有明显的抗肿瘤作用,其机制可能是通过增强小鼠的免疫调节功能来实现的。
Objective: To study the antitumor effect of alkaloids from Oxytropis falcata in mice with further inquiry into the possible mechanism.Methods: The S180 bearing mice were prepared and treated with NS and total alkaloids by ig,with CTX by ip.After 10 days' administration,body weight,tumor weight,thymus index and spleen index of the mice were observed and calculated.Results: The tumor inhibition rates of high,moderate and low dosage of total alkaloids were 74.8%,79.3% and 69.4%.The thymus index and spleen index in all dosages were higher than those in positive group and NS group with no significant difference.Conclusion: The total alkaloids from Oxytropis falcata had significant antitumor effect and the mechanism was probably enhancing the immune function of mice.
