目的研究藏药镰形棘豆对SMMC-7721人肝癌细胞的凋亡及Bcl-2和Bax蛋白表达的影响。方法镰形棘豆总黄酮不同剂量作用于SMMC-7721细胞后,AnnexinV—FITC/PI双染法检测SMMC-7721细胞的凋亡;蛋白质印迹法检测对凋亡相关蛋白Bcl-2和Bax表达的影响。结果镰形棘豆总黄酮作用SMMC-7721细胞后可观察到凋亡细胞的比例明显升高;Bcl-2蛋白表达下降,Bax蛋白表达与对照组相比虽有所降低,但Bcl-2/Bax比值显著减小。结论镰形棘豆总黄酮可明显抑制SMMC-7721细胞的增殖,并诱导细胞凋亡,其分子机制可能与下调Bcl-2基因表达和降低Bcl-2/Bax的比值有关。
OBJECTIVE To probe the influence of Oxytropis falcata on apoptosis and expression of Bcl-2 and Bax proteins in SMMC-7721 cells. METHODS Annexin V-FITC/PI assay was applied to detect apoptosis in SMMC-7721 cells after being treated with TFOF ( total flavonoids of O. falcata). Changes of Bcl-2 and Bax expression were investigated by Western blot. RESULTS Cell apoptosis was induced significantly in SMMC-7721 cells when subjected to TFOF. Western blot showed that Bcl-2 expression was down- regulated. Although Bax expression was also downregulated slightly comparing with that of control, the ratio of Bcl-2/Bax decreased significantly after TFOF treatment. CONCLUSION The results suggest that TFOF of O. falcata may be a pro-apoptotic agent for he- patic cancer ceils via its modulation of Bcl-2/Bax balance.
