目的:探讨半边旗活性成分Ent-11α-hydroxy-15-oxo-kaur-16-en-19-oic-acid(5F)对C26结肠癌细胞环磷酸腺苷(cAMP)水平的影响。方法:以不同时间、不同浓度5F处理C26细胞,放免法测定cAMP含量。磷酸二酯酶抑制剂3-Isobutyl-1-methylxanthine(IBMX)预先孵育后再与5F联合应用,放免法检测cAMP含量,MTT法检测C26细胞活力。结果:5F可以显著增加cAMP含量,具有时间和剂量依赖性;5F对cAMP含量的作用不受IBMX影响,IBMX可以增强5F对cAMP的促进作用和对C26细胞增殖的抑制作用。结论:5F可能通过激活腺苷酸环化酶(AC)增加C26细胞cAMP含量,这可能是5F抗肿瘤的作用机制之一,5F与IBMX联合应用具有协同效应。
Objective:To investigate the effect of ent-11α-hydroxy-15-oxo-kaur-16-en-19-oic-acid(5F),a compound isolated from Pteris semipinnata L(PsL),on cAMP levels in C26 color cancer cells.Methods:C26 cells were treated with 5F(0~40μg/mL)for different time periods.The cAMP levels were examined by radioimmunoassay.IBMX was used for pre-incubation and then combined with 5F,cAMP levels were measured.Viability was examined using a MTT method.Results:5F increased cAMP levels significantly,in a concentration and time dependent manner.The effect was not affected by IBMX.IBMX can enhance the role of 5F on cAMP levels and the inhibition of C26 cell proliferation.Conclusion:5F may increase the cAMP content of C26 cells by activating the adenylate cyclase(AC),which may be one of the anti-tumor mechanisms of 5F.5F has a synergistic effect with IBMX.
