目的采用重组类人胶原蛋白Ⅱ(recombinant human-like collagenⅡ,RHLCⅡ)制备仿生人工骨复合材料,并对复合材料进行研究。方法采用相分离-冷冻干燥法制备RHLCⅡ复合材料;利用扫描电镜、透射电镜、X射线衍射仪、红外光谱仪和万能电子材料实验机对材料的理化性能进行表征;选择急性毒性实验、溶血实验、皮内实验和热原实验对材料的生物相容性进行评价。结果RHLCⅡ复合材料在晶相组成、化学组成和晶体尺寸方面均类似于天然骨,抗压强度和弹性模量介于致密骨和牙本质之间;复合材料的急性毒性实验、溶血实验、皮内实验和热原实验均为阴性。结论RHLCⅡ复合材料具有良好的理化性质和生物相容性,有望成为培养自体成骨细胞的理想支架材料。
Aim To develop an artificial bone-recombinant human-like collagen Ⅱ(RHLCⅡ) composite material and study it.Methods RHLCⅡ composite materials were developed by phase separation-freeze drying method;micro-morphology,structure,crystallite size,crystallite phase and chemical composition of the material were characterized by SEM,TEM,XRD and FTIR.The compressive strength of the material was measured using a electronic universal-testing machine.The biocompatibility experiments included acute systemic toxicity test,hemolytic test,intracutaneous test and pyrogenic reaction test.Results Crystallite phase,chemical composition and crystallite size of RHLCⅡ composite material developed by phase separation-freeze drying method were similar to that of natural bone.Compressive strength and modulus of elasticity of the material were higher than that of cortical bone and lower than that of dentine.The material presented negative results in those biocompatibility experiments.Conclusion RHLCⅡ composite materials have good physical and chemical properties and good biocompatibility,therefore,it is expected that the material will be ideal for the transplantation of autogenous osteoblasts to regenerate bone tissue.
