研究重组类人胶原蛋白Ⅱ(RHLCⅡ)在CM52树脂上的吸附平衡,考察pH值、NaCl浓度及温度对吸附平衡的影响。结果表明:当4.0〈pH≤5.0时,随着pH值增大,吸附容量减小,解离系数虬增大;当3.0≤pH〈4.0时,随着pH值增大,吸附容量增大,解离系数蚝减小;随着NaCl浓度增大,吸附容量减小,解离系数民增大;温度对Langmuir方程参数的影响较小,温度每升高10℃,吸附容量增加3%左右,解离系数K的变化不显著。RHLCⅡ在CM52树脂上的吸附基本符合Langmuir方程,为离子交换法规模制备RHLCⅡ提供了理论依据。
The ion exchange adsorption equilibrium for Recombinant Human-Like CollagenⅡ ( RHLC Ⅱ ) onto resin CM52 was investigated under different pH values, NaCl concentrations and temperatures. The results show that the dissociation constant Kd is higher and the binding capacity is lower when the pH value increases within 4.0 〈 pH ≤ 5.0. However, when pH value increases within 3.0 ≤pH 〈 4.0, Ka is lower and the binding capacity is higher. The binding capacity decreases and Ka increases with the increasing of NaCl concentration. Temprature has less influence on Langmuir model parameters, and higher temperature leads to higher binding capacity, increased by 3% per 10℃ , while no obvious change is observed on Kd. The adsorption of RHLC Ⅱ onto CM52 obeys Langmuir model, which provides theoretical gist for the large scale production of RHLC Ⅱ by ion exchange process.
