目的:观察正肝方对黄曲霉毒素B1(AFB1)诱发的大鼠癌前病变肝组织增殖细胞核抗原(PCNA)和甲胎蛋白(AFP)的影响。方法:100只Wistar大鼠随机分为4组,模型组、正肝方小剂量预防组、正肝方大剂量预防组各30只,正常组10只。除正常组外,先后用AFB,和2-乙酰氨基芴(2-AAF)对3组大鼠进行造模。造模期间,将不同浓度正肝方水剂(0.6g/ml,0.3g/ml)分别灌喂正肝方预防组大鼠;用无菌蒸馏水灌喂模型组大鼠。8周后处死大鼠,肝组织取材。采用免疫组化染色法检测大鼠肝组织PCNA、AFP水平。结果:模型组大鼠肝组织PCNA、AFP染色标记指数(LI)、阳性单位(PU)值最高,正肝方大剂量组大鼠PCNA和AFP染色的Pu值、LI值均较模型组显著降低(P〈0.01或P〈0.05)。结论:大剂量正肝方能有效降低大鼠癌前病变肝组织PCNA和AFP的表达,减轻大鼠癌前病变肝组织的异常增生。
Objective: To observe the effects of Zhengganfang (ZGF) on AFB1 -induced PCNA and AFP in rat hepatic precancerous lesion tissue. Methods: One hundred rats were divided into four groups: control group of model, group of ZGF low dosage, group of ZGF high dosage and control group of normal rats. Except the control group of normal rats, all rats in each group were treated with AFB1 and 2-AAF successively for making model. During the period of making model, the rats in group of ZGF low dosage and group of ZGF high dosage were fed with ZGF at different dosages. The experiment was ended after eight weeks, and the autopsied liver tissues from each rat were checked for PCNA and AFP by immunohistochemistry method. Results: Both the numeral PU and LI of PCNA and AFP in control group of model were the highest. Both the numeral PU and LI of PCNA and AFP in group of ZGF high dosage were less than that of control group of model ( P 〈 0. 01 or P 〈 0. 05 ) . Conclusion: ZGF can effectively inhibite the expression of PCNA and AFP in rat hepatic precancerous lesion tissue, and ZGF has the function of lessening the paraplasm of rat hepatic precancerous lesion tissue.
