本文采用菌落形态观察和分子生物学相结合的方法,对11个菌株进行鉴定。并通过土生空团菌菌株筛选及其与油松幼苗合成试验,探讨土生空团菌菌根形态及其对宿主的作用。结果表明:(1)从菌落颜色形态特征和rDNAITS序列的Blast比对结果来看,本文所选的11个菌株均为土生空团菌的菌种。(2)优良土生空团菌与油松幼苗的根系共生,形成典型的外生菌根,菌根感染率达20.1%,而对照苗无菌根感染现象。(3)土生空团菌对油松幼苗生长的影响:菌根苗总生物量比对照提高186.7%;叶干重增加152%;根干重增加294.3%;根茎比提高189.4%;苗高增加56.5%;侧根数增多166.7%,对促进油松幼苗生长作用明显。
11 bacterial strain were identified and classified by using bacterial colony morphology observation and molecular biology.Mycorrhiza morphology of Cenococcum geophilum and its effect on parasitifer were discussed in this paper.The results show that:(1) According to the Results of colour 、 morphology of bacterial strain and rDNA ITS sequence comparison,all of the 11 bacterial strain selected in this paper were bacterial strain of Cenococcum geophilum.(2) Typical exogenesis mycorrhiza were formed through symbiosis of Fine Cenococcum geophilum and root system of pinus tabulaeforsis sapling,and mycorrhiza infected rate was 20.1%,while mycorrhiza infected phenomenon didn't take place on CK.(3) The impact of Cenococcum geophilum on the growth of Pinus tabulaeformis seedling,compared with CK,its biomass increased 186.7%;dry weight of leaves gained 294.3%;ratio of root and stem increased 189.4%;height of seedling increased 56.5%;amount of lateral root increased 166.7%;it played an obvious role in growth of Pinus tabulaeforsis seedling.
