目的 建立纯度较高的适于实验研究的人蜕膜细胞。方法 胶原酶Ⅰ消化法培养人蜕膜细胞,光镜、扫描电镜、透射电镜观察细胞内部结构,间接免疫染色进行细胞鉴定。结果 倒置显微镜下,可见细胞呈不规则多角形,呈片状铺展生长,核大卵圆形,胞质透明、糖原丰富,内质网扩张明显。免疫组化法检测细胞功能性指标胎盘催乳素,有99%培养的蜕膜细胞呈阳性。结论 该法简便易行,可获得合乎实验要求的人蜕膜细胞,为进一步开展实验研究建立细胞模型。
Objective To establish highly purified functional human decidual cells from human placental in vitro. Methods Collagenase type Ⅰ dispersion method was used to isolate the decidual cells by immunohistochemistry. The structural features were revealed by light microscope, scanning electron microscope, transmission electron microscope. Results Light microscopic examination revealed patchy spreading and irregularpolygonal cells with big and oviform nuclear. The transmission electron microscope examination revealed the cells with abundance and transparent cytoplasm, glycogen granule and endocytoplasmic reticulum. PRL immunohistochemistry assay was adopted to testify the components of the cultured cells. 99 % of the decidual cells was positive for PRL. Conclusions Decidual cells may be obtained with the simple method. We gained pure decidual cells with the part function of endocrine secretion that could offer a model amenable to experimental manipulation.
