western blot是生命科学研究中分析特定蛋白质的常用手段。在分子生物学实验课程已经建立的克隆小鼠GAPDH基因编码序列教学过程的基础上,以IPTG诱导后的原核细胞裂解液为实验材料,建立了适合课题教学的快速斑点印渍检测方法,使学生能够很好地掌握western blot的实验原理和方法;直观地观察到自己整个分子克隆实验的成果,激发了学生的学习兴趣,促进了学生的科学研究能力的提升。
The western blot is a widely used method to detect specific proteins in a sample of tissue homogenate or extract in life sciences study. In succession to the cloning of mouse GAPDH gene in the previous course of molecular biology experiment,the transformed E. coli stain was induced by IPTG and the cell lysate was used as a start material for the experimental teaching. The expression of cloned gene was analyzed by rapid dot blot with a specific antibody. Through the class,the student can learn the technique of western blot,observed the final outcome of their molecular cloning experiment directly,their enthusiasm in learning was stimulated and their scientific researching ability were promoted.
