目的观察体外培养的背角小胶质细胞P2Y受体激活对其[Ca2+]i的影响。方法培养并纯化脊髓背角小胶质细胞,采用激光共聚焦技术观察P2Y受体激活后小胶质细胞[ca2+]i的变化。结果P2Y受体激动剂2-mesADP在10μm即可引起小胶质细胞[ca2+]i升高,当随着2-mesADP浓度增大,小胶质细胞[ca2+]i升高更为明显。结论体外培养的大鼠脊髓背角小胶质细胞P2Y受体激活可以促进细胞[ca2+]i升高。
Objective To investigate the effect of 2 -mesADP (P2Y receptor agonist) on the change of [ Ca2+ ]i in dorsal horn microglial cells. Methods The expression of ox -42 in cultured dorsal horn mi- croglial cells was observed by immunohistochemical staining. 2 - mesADP - induced Ca2 + mobilization in cultured dorsal horn microglia was measured by laser scanning confocal microscopy. Results Cultured dorsal horn microglial cells expressed high level of ox - 42. Furthermore, 2 - mesADP could induce the increase of the Ca2+ fluorescent intensity in microglial ceils in a dose - dependent manner. Conclusion P2Y receptor agonists 2 - mesADP can induce the increase of [ Ca2 + ] i in dorsal horn microglial cells.
