目的对中药复方海藻制剂水煎液各相的体外免疫调节、细胞毒和抗氧化作用进行初步研究。方法采用中性红法测定对小鼠腹腔巨噬细胞吞噬能力的影响;采用MTT法测定对小鼠脾淋巴细胞的增殖作用;采用MTT法测定对肿瘤细胞株的细胞毒活性;采用比色法测定对DPPH自由基、羟基自由基清除能力以及对抗脂质过氧化能力、还原能力和总体抗氧化能力的影响。结果复方海藻制剂的乙酸乙酯相与正丁醇相显示中等强度的免疫调节活性.乙酸乙酯相显示强抗氧化活性。结论免疫调节与抗氧化作用可能是中药复方海藻制剂抗肿瘤作用的药理基础。
Objective To study the immunomodulatory activities, cytotoxic activities and antioxidant activities of different fractions from traditional Chinese medicine seaweed complex prescription (SCP) in vitro. Methods The effects on phagocytosis of mouse macrophages were evaluated using the Neutral Red Dye method. The effects on the proliferation of mouse lymphoeytes and the cytotoxic activities against tumor cell lines were assayed by MTT method. The antioxidant activities, including DPPH free radical scavenging activity, hydroxyl radical scavenging activity, inhibition of lipid peroxidation, reducing power and total antioxidant activity, were detected using different methods in vitro. Results The ethyl acetate fraction and n-BuOH fractions of SCP had moderate immunomodulatory activities, and the ethyl acetate fraction showed strong antioxidant activities. Conclusion Immunomodulatory activities and antioxidant activities might be the effective ways of antitumor effects of SCP.
