目的:考察不同剂量参麦注射液(SMI)对心衰犬血清肿瘤坏死因子α(TNF-α),白介素1β(IL-1β),白介素6(IL-6)的影响。方法:将造模成功4周后的心衰犬20只随机分为模型对照组和SMI低、中、高剂量组,每组5只。模型对照组予生理盐水20mL,低、中、高剂量组分别予参麦注射液0.517,1.034,1.551ml·kg^-1(生理盐水稀释至20mL)ivqd×7。分别于给药前及给药后72h,144h时点取静脉血2mL冰冻保存,用放射免疫分析法测定血清TNF-α,IL-1β,IL-6浓度。结果:SMI低、中、高剂量组心衰犬血清TNF-α,IL-1β,IL-6水平较模型对照组下调。结论:SMI能使心衰犬血中TNF-α,IL-1β,IL-6水平下调,并呈现一定的量效、时效关系。
Objective:To investigate effect of different dose Shenmai Injection(SMI) on blood concentration of TNF-α,IL-1β,IL-6. Method:Twenty model dogs with heart failure(HF) were randomly divided into 4 groups:the control one(0. 9% NaCl injection 20 mL iv qd × 7,n = 5) and the low,middle and high dose ones of SMI(administrated by 0. 517,1. 034 and 1. 551 mL·kg^-1,diluted to 20 mL with 0. 9% NaCl injection,iv qd × 7,n = 5) . Blood samples(2 mL) were collected before administration and at 72 h and 144 h after that,refrigerated(at-80 ℃) to be determined for the blood concentration of TNF-α,IL-1β,IL-6 by radio-immunity assay. Results:Blood concentration of TNF-α,IL-1β,IL-6 in the different dose SMI groups was higher than that in the control one,and it showed some dose-effect relationship. Conclusions:SMI could effectively reduce blood concentration of TNF-α,IL-1β,IL-6 in the model dogs with heart failure and showed some dose-effect and time-effect relationships.
