目的:参麦注射液对心衰犬地高辛肾排泄的影响。方法:将造模成功4周后的心衰犬20只随机分为对照组和参麦注射液低、中、高剂量组,每组5只。予地高辛注射液7.41μg/kgivst后,立即予对照组生理盐水20mlivqd×5,低、中、高剂量组分别予参麦注射液0.517、1.034、1.551ml/kg(均用生理盐水稀释至20ml)ivqd×5。于给药后24h、48h、72h、96h收集每日尿液计量,留取尿样4ml;用高效液相色谱-质谱联用法检测尿液地高辛浓度。结果:与对照组及3种剂量参麦注射液之间比较,对地高辛的肾排泄均未见显著影响,但低剂量组的尿地高辛累积排泄量与累积排泄率有高于对照组和中、高剂量组的趋势(P〉0.05)。结论:参麦注射液对地高辛肾排泄无明显影响。
Objective:To clarify the effect of Shenmai Injection(SMI)on digoxin (DG) urinary excretion in model dogs with heart failure.Method:Twenty model dogs with HF were randomly divided into 4 groups:the control one and the low,middle and high dose ones of SMI (n=5).After digoxin injection 7.41 μg/kg iv st,the control group was administrated with 0.9% NaCl injection 20ml iv qd ×5d,and the 3 trial ones of different dose were done with SMI 0.517,1.034 and 1.551 ml/kg,diluted to 20ml with 0.9% NaCl injection,iv qd ×5d.The urine samples were collected at 24h,48h,72h and 96h after the administration.The digoxin concentration of urine was determined by LC/MS/MS.Result:The four groups had no significant difference in urinary excretion amount,accumulative urinary excretion amount,urinary excretion rate and accumulative urinary excretion rate.The low dose group showed only a tendency of increasing the accumulative urinary excretion amount and accumulative urinary excretion rate(P0.05).Conclusion:SMI has no significantly effect on DG urinary excretion.
