目的探讨增殖抑制基因(HSG)在乳腺癌中的表达及其与增殖细胞核抗原(PCNA)和癌基因P21WAF1的关系。方法采用免疫组化法检测63例乳腺癌及20例癌旁正常乳腺标本中HSG、PCNA和P21WAF1的表达。结果乳腺癌组HSG表达率较癌旁正常乳腺组下降,PCNA和P21WAF1表达率较癌旁正常乳腺组升高。乳腺癌组HSG低表达与组织学分级、淋巴结转移和ER有关,PCNA和P21WAF1高表达与组织学分级、淋巴结转移、复发和预后有关,HSG与PCNA的表达呈负相关。结论HSG低表达和PCNA、P21WAF1高表达均与乳腺癌细胞的增殖和乳腺癌的发生、发展有关,是判断乳腺癌生物学行为的良好指标。
Objective To determine the relationship between the expression of novel hyperplasia suppressor gene and PCNA and P21WAF1 in breast cancer tissues. Methods The expression of HSG, PCNA and P21WAF1 were detected by immunohistochemical method in 63 breast cancer tissues and 20 samples paracancer normal breast tissues. Results The expression rate of HSG in the breast cancer tissues was lower than that in the paracancer normal breast tissues. The expression rate of PCNA and P21WAF1 in the breast cancer tissues was higher than that in the paracancer normal breast tissues. The lower expression of HSG in breast cancer was correlated with histological grade, lymph node metastasis and ER, The higher expression of PCNA and P21WAF1 was related with histological grade, lymphnode metastasis, recurrence and prognosis. The expression of HSG was negatively correlated with the expression of PCNA in breast cancer tissues. Conclusions In breast cancer tissues ,the downregulation of HSG and uprcgulation PCNA and P21WAF1 may have a close relationship with the hyperplasia of breast cancer cell and may be marker of the occurrence and development of breast cancer.
