目的:探讨线粒体融合蛋白2(mitofusin 2,Mfn2)在非小细胞肺癌(non-small-cell lung cancer,NSCLC)组织和非癌肺组织中的表达及与临床病理特征之间的关系。方法:应用原位杂交和免疫组织化学法检测92例NSCLC组织和27例非癌肺组织中Mfn2 mRNA及蛋白的表达。结果:肺鳞癌组织中Mfn2 mRNA和蛋白的阳性表达率(83.33%和89.58%)均高于肺腺癌(56.82%和65.91%),且2者均高于非癌肺组织的阳性表达率(25.93%和29.63%),3组间比较差异有统计学意义(P〈0.001和P〈0.01)。高分化和中分化的肺癌组织中Mfn2 mRNA和蛋白的阳性表达率(93.75%和100%,91.67%和91.67%)均高于低分化的肺癌组织(21.43%和42.86%),差异有统计学意义(P〈0.001)。Mfn2 mRNA和蛋白的表达与患者的性别、年龄、肿瘤大小、TNM分期和淋巴结转移无关(P〉0.05)。肺癌组织中Mfn2 mRNA和蛋白的表达具有较好的一致性。结论:Mfn2参与了肺癌的发生和发展,Mfn2的表达与肺癌的组织学类型及分化程度有关。
Objective:To investigate the expressions of Mfn2(mitofusin 2) in non-small-cell lung cancer(NSCLC) tissues and non-cancerous lung tissues,and analyze its relationship with clinicopathological characteristics of lung carcinomas.Methods:The expressions of Mfn2 mRNA and protein in 92 cases of NSCLC tissues and 27 cases of non-cancerous lung tissues were detected by in situ hybridization and immunohistochemical methods.Results:The positive rates of Mfn2 mRNA and protein in pulmonary squamous cell carcinoma were higher than those in adenocarcinoma(83.33% and 89.58% vs 56.82% and 65.91%),respectively.The positive rates of Mfn2 mRNA and protein in NSCLC were higher than those in the non-cancerous lung tissues(25.93% and 29.63%).The difference was statistically significant among the three groups(P0.001 and P0.01).The expressions of Mfn2 mRNA and protein in well-differentiated(93.75% and 100%) and moderately-differentiated NSCLC(91.67% and 91.67%) were higher than those in poor-diffe-rentiated NSCLC(21.43% and 42.86%).The difference was significant(P0.001).The expressions of Mfn2 mRNA and protein had no correlation with the gender,age,tumor size,TNM stage and lymph node metastasis(P0.05).The expression of Mfn2 mRNA was consistent with that of Mfn2 protein in NSCLC.Conclusion:Mfn2 was involved in the initiation and progression of lung cancer,and the expression of Mfn2 was related to the histological types of lung cancer and its differentiation degree.
