目的探讨颗粒细胞在卵母细胞体外成熟(in vitro matueation,IVM)过程中发挥的作用。方法选取8-12周龄的CD-1雌鼠,取卵前48h腹腔注射HMG。取卵日处死小鼠,开腹取卵巢,于卵巢表面穿刺获得GV期卵母细胞。将所得的卵母细胞随机分成三组:A组保留颗粒细胞,B组为混合培养组,将一半的卵母细胞脱去颗粒形成裸卵与另一半的颗粒细胞卵母细胞复合体(Cumulus-oocyte comeplexes,COCs)共同培养,本组只观察DOs部分,C组全部脱去颗粒细胞。将各组卵母细胞置于含FSH(75IU/L)和HCG(150 IU/L)的G1中培养24h后,对成熟卵行单精子显微注射。观察各组间卵母细胞的成熟率及受精后胚胎发育情况。结果 1三组间成熟率比较差异无统计学意义(P〉0.05),但A、B组略高。2A、B组优质胚胎率明显高于C组(P〈0.05),且A组与B组间的差异无统计学意义(P〉0.05)。结论颗粒细胞在卵母细胞成熟过程中发挥着重要作用:1单独对DOs体外成熟,后期发育不佳。2DOs与COCs共培养效果较好。
Objective To investigate the role of granulosa cells during in-vitro maturation of oocytes.Methods CD-1female mice of 8-12 weeks were selected,48 hours before the eggs pick up celiac injection HMG.Put to death in mice and took eggs in ovary,laparotomy in ovarian surface piercing GV stage oocytes.The oocytes obtained were randomly divided into three groups:group A reservation granulosa cells,group B of mixed culture,will be half of the oocyte remove particles formed eggs and the other half naked granulosa cells oocyte complexes(Cumulus oocyte- comeplexes,COCs)jointed training,only observe the DOs,group C all took off granulosa cells.Put each oocyte in FSH(75IU/L)and HCG(150IU/L)in the G1 training after 24 hours,single sperm microinjection of mature eggs.Oocyte maturation rate was recorded among three observation group after fertilization and embryonic development.Results The mature rate had no obvious difference among the three groups(P〉0.05),but it was slightly higher in group A and group B.The rate of high quality embryo was obviously higher in group A and group B than that of group C(P〈0.05),and the differences between group A and group B had no statistical significance(P〉0.05).Conclusion Granular cell in the oocyte maturation plays an important role in the process on in vitro maturation of DOs alone and in the late development.The effect of DOs with COCs co-culture is better.
