[目的]建立从冷冻血凝块中提取基因组DNA的改良方法.[方法]取冷冻血凝块在4℃条件下解冻,经高盐及高EDTA提取液处理,消化液消化,异丙醇抽提.[结果]用改良异丙醇法成功地从人血凝块中提取基因组DNA,其质量浓度为(22.5±7.2)mg/L,光吸收比值A260/A280为1.63~2.00.[结论]应用改良法提取的基因组DNA的总量较多,提取率高,PCR扩增效果较好,可用于血凝块基因组DNA的提取.
OBJECTIVE To explore a simple method of DNA extraction from human blood clots. METHODS Blood clot was thawed at 4℃, homogenized extraction buffer including higher sodium chloride and higher EDTA, the homogenized samples were digested with digestion buffer and extracted with phenol-chloroform. RESULTS The genomic DNA was successfully extracted. The concentration of the extracted DNA was (22.5 % 7.2) mg/L, and OD value A260/280 was 1.63 % 2.00. CONCLUSION The improved extraction method is reliable for obtaining high quantities of DNA from blood clot and suites for PCR amplification.
