目的观察蛋白激酶A(PKA)信号通路在食管癌EC9706细胞增殖和凋亡中的作用。方法利用PKA特异性阻断剂H89作用于食管癌EC9706细胞,倒置显微镜下观察食管癌EC9706细胞形态学改变;WST-8检测细胞增殖情况;Annexin V-FITC和PI双染法检测细胞凋亡。结果与对照组相比,H89作用于食管癌EC9706细胞24 h后,部分细胞出现细胞形态缩小,细胞增殖减慢;细胞增殖实验显示,吸光度值降低(P〈0.05),细胞存活率下降;细胞凋亡检测结果显示,阻断PKA通路,促进细胞早期凋亡和晚期凋亡(P〈0.05)。结论利用H89阻断PKA信号通路,可以改变食管癌EC9706细胞生物学特性,抑制细胞增殖,促进细胞凋亡。
Objective To observe the function of protein kinase A(PKA) signaling pathway on cell proliferation and apoptosis of esophageal cancer cell line EC9706.Methods EC9706 cells were treated with the specific PKA inhibitor H89.Morphological changes of EC9706 cells were observed with inverted microscope;cell proliferation was measured with WST-8;cell apoptosis of EC9706 were measured with Annexin V-FITC and PI double staining.Results Compared with the control group,after the 24 h treatment with H89,some cells showed morphology changes,cell proliferation slowed down;cell proliferation experiments revealed that the absorbance decreased(P〈0.05) and cell survival rate also decreased;the cell apoptosis results showed that blocking the PKA pathway,promoting early apoptosis and late apoptosis(P〈0.05).Conclusion H89 can alter the biological characteristics of EC9706 cells,inhibit proliferation and promote apoptosis.
