目的观察不同浓度的互隔交链孢霉素(altenuene,ALT)对小鼠胚胎成纤维细胞(NIH/3T3)中cAMP水平的影响。方法以NIH/3T3为研究对象,观察以0、2.5、5、10、15和20μmol/L ALT作用30min,并设0.4mmol/L烷化剂甲璜酸甲酯(methyl methanesulfonate,MMS)为阳性参照物,使用超声法破碎细胞,以^125I标记的放射免疫分析(RIA)检测细胞内cAMP水平。结果与对照组相比,5、10、15和20μmol/L ALT能升高细胞内cAMP水平(P〈0.05);而以0.4mmol/L MMS处理细胞,cAMP水平同样升高。结论ALT对细胞具有明显的影响,可能与经由第二信使cAMP所介导的相关信号通路有关。
Objective To observe the effects of Ahenuene of different concentrations on cAMP in NIH/3T3 cell line. Methods NIH/3T3 cells were cultured for 30 rain in the medium supplemented with ALT at 0, 2.5, 5, 10, 15, 20 μmol/L and MMS at 0.4 mmol/L for positive control. Then the level of cAMP in NIH/3T3 cell was observed by ^125I-cAMP RIA. Results After NIH/3T3 cells were exposed to different concentrations of ALT for 30 min, the cellular level of cAMP was significantly increased in a concentration-dependent, and in positive control group the level of cAMP increased the same (P 〈 0.05 ). Conclusion ALT can increase the concentrations on cAMP in NIH/3T3 cells, which related to the single pathway via the second messenger cAMP possibly.
