基于肌红蛋白-硅酸铝(Mb-AlSiO4)凝胶膜修饰电极,制备了一种非媒介体型的过氧化苯甲酰(BPO)生物传感器,并用于小麦粉中BPO的直接快速检测。用扫描电子显微镜(SEM)、X-光电子能谱(XPS)和原子力显微镜(AFM)对AlSiO4凝胶修饰电极进行了表征。电极表面的Mb-AlSiO4凝胶膜有均匀的多孔结构,能长时间保持肌红蛋白活性。该传感器对BPO的电化学还原表现出良好的催化活性,BOP还原峰电流与其浓度在1×10^-6~4×10^-4mol/L的范围内有良好的线性关系,相关系数为0.997,检测限是5×10^-7mol/L。具有灵敏、稳定、易于制备的优点,适合于样品的现场快速测定,此技术平台可推广到粮油食品中其它添加剂的检测。
An unmediated hydrogen peroxide sensor was designed by employing a myoglobin-AlSiO4 sol- gel film modified electrode. The myoglobin exhibited direct (unmediated) electrochemistry at the modified electrode. Both Mb and Mb entrapped AlSiO4 sol- gel films were characterized in detail by scanning electron microscopy (SEM), Fourier transformation infrared spectroscopy (AFM), X - ray photoelectron spectroscopy (XPS) and electrochemical techniques. Results show: The sol - gel films are uniform porous and show a very low mass transport barrier and a regular dense. Mb -AlSiO4 sol- gel matrix retains the native structure and activity of entrapped Mb and prevents the enzyme from leaking out of the film. The protein also shows elegant catalytic activity towards the electrochemical reduction of benzoyl peroxide (BPO). Consequently, a prototype BPO sensor is prepared. Under optimum conditions, this sensor provides a linear response over the BPO concentrations in the range of 1×10^-6- 4×10^-4mol/L, correlation coefficient 0.997. The detection limit is 5×10^-7 mol/L. This configuration is shown to be sensitive, stable and easy to be fabricated.
