目的：研究健脾解毒方诱导胃癌细胞凋亡的作用并探讨其部分机制。方法：建立裸鼠人胃癌MKN-45细胞皮下移植瘤模型,随机分为空白对照组、替加氟组、健脾解毒方组、联合用药组4组,每组12只;给药14 d后观察各组对裸鼠胃癌的治疗作用及其对荷瘤裸鼠生存期的影响;TUNEL法检测各组胃癌细胞凋亡情况;分别采用实时荧光定量PCR方法和免疫组化S-P法检测健脾解毒方干预后Bcl-2、Bax的mRNA和蛋白表达情况。结果：与空白对照组相比,健脾解毒方组凋亡指数显著升高、Bcl-2 mRNA水平及Bcl-2蛋白表达显著降低、Bax的mRNA和蛋白表达明显上调、Bcl-2/Bax显著下降,健脾解毒方组和联合用药组均能明显抑制瘤体生长（P〈0.01）,延长荷瘤鼠的生存期（P〈0.01）。联合用药组在抑制胃癌细胞生长和诱导胃癌细胞凋亡方面的作用均优于单纯化疗药组和单纯健脾解毒方组（P〈0.05或P〈0.01）。结论：健脾解毒方能抑制裸鼠胃癌细胞生长,诱导胃癌细胞凋亡,其机制可能与调控Bcl-2、Bax表达有关。

英文摘要：

Objective：To discover possible mechanisms of induction of apoptosis of Jianpi Jiedu Decoction on MKN-45 nude mice model.Methods：Experimental MKN-45 nude mice were randomly divided into 4 groups：NS control group,Ft-207 group,Jianpi Jiedu therapy group（JPJD）and combination therapy group（JPJD＋ Ft-207）.14 d after treatment,the tu-mor growth and the survival time of the mice in each group were observed;the apoptosis indexes were examined with TUNEL;The expression of Bcl-2 mRNA and Bax mRNA were detected with QRealTime-PCR;The protein of Bcl-2 and Bax were examined with S-P immunohistochemical technique.Results：Compared to NS control group,apoptosis indexes was significantly increased,the protein and mRNA expression of Bcl-2 were down-regulated,the mRNA and protein ex-pression of Bax were up-regulated,and the Bcl-2/Bax were reduced in JPJD group.Tumor weights were decreased,and the survival time of the nude mice in JPJD group and combination therapy group was extended（P0.01）.But all the items above were more obviously in combination therapy group.Conclusions：Jianpi Jiedu Decoction can significantly inhibit the growth of MKN-45 cells.The mechanism of Jianpi Jiedu Decoction for inducing cells apoptosis is related to regulation of Bcl-2 and Bax expressions.
