【目的】鉴定和比较砂梨品种‘黄花’(Pyrus pyrifolia Nakai‘Huanghua’)及其芽变‘绿黄花’(Pyrus pyrifolia Nakai‘Lühuanghua’)成熟期果皮代谢产物的差异。【方法】利用气相色谱-质谱联用(GC-MS)和液相色谱-质谱联用(LCMS)技术,对2者成熟期果皮中代谢产物进行鉴定。同时,对这2种梨果皮中代谢物质的峰响应强度数据进行主成分分析(PCA)和偏最小二乘法分析(PLS-DA)。【结果】2者果皮中代谢物的差异使得梨果皮色泽发生变异。GC-MS的结果中共得到18种差异性代谢产物,主要包括草酸、苹果酸和柠檬酸等有机酸类,丙氨酸和L-天门冬酰胺等氨基酸类以及果糖、葡萄糖和蔗糖等糖类物质;LC-MS的结果中共得到43种差异性代谢产物,主要包括顺式-15-二十四碳烯酸、5,9-二十四碳二烯酸、亚油酸、亚麻酸、硬脂酸、二十二烷酸和二十四烷酸等脂肪酸类,邻苯二甲酸二己基酯和2-单亚油酰甘油酯等酯类,熊果酸和角鲨烯等三萜类,5-氧-反式-对-香豆酰-奎宁酸、香草酸、熊果苷和绿原酸等酚类,黄酮类,松柏醛和肉桂酸等苯丙烷类,豆甾醇等甾醇类以及δ-生育三烯酚和α-生育三烯酚等维生素E类物质。【结论】‘黄花’与‘绿黄花’果皮色泽上的不同很可能是这些差异性代谢产物共同作用的结果。
[Objective] The aim of this study was to discover differential metabolites from the skin of mature fruit in ' Huanghua' and its bud mutant ' Luhuanghua' (Pyrus pyrifolia Nakai). [Methods] The metabolites in skin were measured by gas chromatography-mass spectrometer (GC-MS) and liquid chromatography-mass spectrometer (LC-MS). The data were analyzed using principal component analysis and partial least squares-discriminate analysis (PLS-DA). [Results] Differences in pear skin color resulted from the significant difference of metabolite. Eighteen differential metabolites were found by GC-MS, mainly including organic acids (Oxalic acid, Malie acid and Citric acid) , amino acids (Alanine and L- Asparagine ) and sugar (D-Fructose, D-Glucose and Sucrose). Forty-three differential metabolites were obtained by LC-MS, primarily including fatty acids (cis-15-tetracosenoic acid, 5,9-tetracosadienoic acid, linoleic acid, linolenic acid, stearic acid, docosanoic acid and tetracosanoic acid), esters (dihexyl 1,2-benzenedicarboxylate and 2-Monolinoleoylglycerol) , triterpenoids (ursolic acid and squalene ), phenolics (5-O-trans-p-coumaroyl quinic acid, vanillic acid, chlorogenic acid and arbutin, ), flavonoids, phenylpropanoids (coniferyl aldehyde and cinnamic acid), sterols (Stigmasterol) and vitamin E (δ- tocotrienol and α- tocotrienol). [ Conclusion] The difference of skin color between ' Huanghua' and its bud mutatant 'Luhuanghua' might be caused by the synergistic reaction of these differential metabolites.
