目的研究复尔康注射液在体外对血管生成的抑制作用及对肿瘤细胞血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法采用细胞计数试剂盒(cell counting kit-8,CCKS)、细胞刮痕法(cell wounding healing assay)和小管形成试验(tube formation assay)等检测复尔康注射液对人微血管内皮细胞株(human microvascular endothelial cells,HMECs)增殖、迁移和小管形成的影响;采用CCK8、免疫组化SABC法检测复尔康注射液对HepG2细胞增殖和VEGF表达的影响。结果复尔康注射液明显抑制HMEC细胞的增殖,IC50为(11.44±3.97)mg·L^-1;复尔康注射液质量浓度在3.125mg·L^-1时,表现出抑制HMEC细胞迁移和小管形成的作用,且随着质量浓度的增加,抑制作用愈明显,呈现出较好的剂量依赖关系;复尔康注射液还能明显抑制HepG2细胞的增殖,减弱VEGF的分泌表达。结论抗血管生成作用可能是复尔康注射液抗肿瘤作用的机制之一。
Objective To study the antiangiogenic activity of Fuerkang ( traditional Chinese medicine) injection in vitro and the effect on the expression of VEGF in tumor cells. Methods The antiangiogenesis activity of Fuerkang injection on human microvascular endothelial cells (HMECs) was investigated by cell counting kit-8 assay( CCK8 ) ,wounding migration assay and matrigel tube formation assay. The cytotoxicity of Fuerkang injection on HepG2 was determined by CCK8 and the expression of VEGF was tested by SABC of immunohistochemical method. Results Fuerkang injection inhibited the proliferation of HMECs, the IC50 value was ( 11.44 ±3.97 ) mg. L^-1. Furthermore, Fuerkang injection strongly inhibited the migration and capillary- like networks development of HMECs in a dose-dependent manner. Fuerkang injection also inhibited the proliferation and the expression of VEGF in HepG2 cells. Condusion Antiangiogenic effect may be one of the mechanisms of antitumor action of Fuerkang injection.
