目的:研究青蒿油制剂复尔康注射液对肝癌HepG2细胞增殖和凋亡的影响。方法:CCK8试剂盒(cell counting kit-8)检测复尔康注射液对肝癌HepG2细胞增殖的抑制作用;Hoechst33258荧光染色观察HepG2细胞凋亡的形态学改变;流式细胞仪AnnexinV/PI双染法检测HepG2细胞的凋亡率。结果:复尔康注射液可抑制HepG2细胞的增殖,IC50值为29.63±1.22μg/ml;复尔康注射液0.3125-10μg/ml作用48h后,Hoechst33258荧光染色均可见HepG2细胞出现核浓缩、核碎裂等细胞凋亡形态学改变,且呈现出一定的剂量依赖性;AnnexinV/PI双染法检测显示,不同浓度的复尔康注射液均可诱导HepG2细胞凋亡,凋亡率分别为4.71%±0.71%、10.05%±0.67%、15.08%±1.16%、23.59%±1.65%、32.30%±1.87%、42.09%±1.43%。结论:复尔康注射液可抑制HepG2细胞的增殖,诱导其凋亡。
Objective: To study the effect of Fuerkang injection on the proliferation and apoptosis of hepatoblastoma G2 (HepG2) cell. Methods: The cytotoxicity of Fuerkang injection on HepG2 was determined by CCK8 ( cell counting kit-8 ) ; The cell apoptosis morphological changes was observed using Hoechst 33258 staining; The rate of apoptosis was detected by flow cytometry with Annexin-V/PI staining. Results: The Fuerkang injection can inhibit the proliferation of HepG2 cells, the IC50 value was 29.63±1. 22μg/ml; Typical morphological changes of cell apoptosis such as nuclear concertration and nuclear fragmentation were observed after treatment of Fuerkang injectiong at concentration of 0.3125 - 10μg/ml for 48 h in a dose-dependent manner; The apoptosis of HepG2 cells was induced by different concentration of Fuerkang injection and the apoptosis rate was 4.71% ± 0.71%, 10.05% ± 0.67%, 15.08% ± 1.16% ,23.59% ± 1.65% ,32.30% ± 1.87% and 42. 09% ± 1.43%, respectively . Conclusion: Fuerkang injection can inhibit the proliferation of HepG2 cells and induce their apeptosis.
