磁流体热疗对小鼠Lewis肺癌治疗作用的实验研究

目的探讨磁流体热疗对小鼠Lewis肺癌的治疗作用。方法将Lewis肺癌的细胞悬液接种于C57/BL6小鼠的皮下,当肿瘤长至直径(0.8±0.1)cm时,将20mg磁流体直接注射到肿瘤内部,24h后在交变磁场下加温(46.0℃,30min),观察磁流体热疗后肿瘤体积的变化、瘤体的病理变化,对肿瘤生长及小鼠生存期的影响。结果加温治疗后14天,实验组肿瘤体积抑制率达81.6%,与对照组比较,瘤体的增长受到明显的抑制(P〈0.05),其中有4只小鼠的瘤体完全消失;肿瘤没有消失小鼠存活(37.1±8.6)d,瘤体消失的小鼠存活超过90d,与对照组比较生存期明显延长(P〈0.01);组织学检查肿瘤细胞呈凋亡、坏死样改变。结论磁流体热疗能有效抑制小鼠Lewis肺癌的生长,延长生存期,为磁流体热疗进一步研究提供实验依据。

Objective To assess the therapeutic effect of magnetic fluid hyperthermia on Lewis lung carcinoma in mice. Methods C57BL/6 mice were subcutaneously inoculated with Lewis lung carcinoma cells. When the tumor grew to (0.8 ±0.1 ) cm in diameter, magnetic fluid (20 rag) was directly injected into the center of the tumor. After 24 hours, the tumor was heated in an alternating magnetic field (46. 0 ℃ ) for 30 minutes. The therapeutic effect was assessed by measuring the tumor volume and pathological examination. Results Fourteen days after hyperthermia, the volume inhibitory rate in the experiment group was 81.6% , which was significantly lower than that in the control ( P 〈 0.05). In 4 of the 12 mice in the experiment group, the tumor disappeared completely. The mice with the tumor survived for (37.1 ± 8.6) days, while those with the tumor disappeared survived for over 90 days ( P 〈 0.01 ). Pathological examinations showed necrosis and apoptosis of the tumor cells. Conclusions Magnetic fluid hyperthermia can inhibit the growth of Lewis lung carcinoma and prolong the survival time of the mice. The results may provide experimental basis for further researches on magnetic fluid hyperthermia.
