目的研究导痰汤是否能够通过对内皮细胞间黏附分子-1(ICAM.1)和p38通路的调节而干预颈动脉硬化。方法新西兰白兔随机分为对照组、模型组和导痰汤大、中、小剂量组(10、8、6g/kg导痰汤灌胃)。通过免疫组化方法,观察导痰汤对颈动脉硬化家兔血管内皮细胞ICAM-1和p38表达的影响。结果模型组血管壁内皮细胞的胞浆中有ICAM-1和p38强阳性表达,显著高于对照组(P〈0.01)。导痰汤治疗组细胞内ICAM-1和p38阳性表达显著低于模型组(P〈0.01),且随剂量的增加,导痰汤对ICAM-1和p38的抑制逐渐增强;而p38活性与ICAM-1水平呈显著正相关(r=0.757,P〈0.01)。结论导痰汤可以有效抑制颈动脉硬化家兔血管内皮细胞中ICAM-1和p38的表达。
Objective To study whether Daotan Tang can interfere carotid atherosclerosis through regulating intercellular adhesion molecular-1 (ICAM-1) and p38 pathway. Methods New Zealand rabbitswere randomly divided into control group, model group and treatment groups with Daotan Tang in different doses [ high-dose group (10 mL/kg), mid-dose group (8 mL/kg) and low-dose group (6 mL/ kg) given intragastrically]. The influences of Daotan Tang on the expressions of ICAM-1 and p38 were observed by using immunohistochemistry technique. Results In model group, the expressions of ICAM- 1 and p38 showed strong positive in endochylema of endothelial cells of vessel wall, which were significantly higher than those in control group (P 〈0.01 ). The positive expressions of ICAM-1 and p38 were significantly lower in treatment groups than those in model group ( P 〈 0. 01 ), and this inhibitory effect increased as the dose of Daotan Tang increased. The activity of p38 was significantly correlated to ICAM-1 level ( r = 0. 757, P 〈 0.01 ). Conclusion Daotan Tang can effectively inhibit the expressions of ICAM-1 and p38 in vessel endothelial cells in rabbits with carotid atherosclerosis.
