目的:初步探索红花黄色素(以下简称SY)对大鼠急性脑缺血后不同时间点炎性损伤的保护作用。方法:健康雄性SD大鼠随机分假手术组、MCAO模型组和SY组。假手术组、模型组和SY组又各分4个时间点(3h,6h,12h,24h)。即假手术组(3h,6h,12h,24h),MCAO模型组(3h,6h,12h,24h),SY组(3h,6h,12h,24h)共12组各15只。术后取眶周血用ELISA法测炎症因子TNF-α、IL-6及IL-10水平,之后断头取脑切片行HE染色分析。结果:SY可抑制脑缺血后促炎因子TNF-α(3 h、24 h)和IL-6(3 h、6 h和24 h)升高的作用,并且促进抗炎因子IL-10含量升高(3 h、12 h和24 h)。SY对大鼠急性脑缺血后24h脑组织切片炎性损伤有明显的缓解作用。结论:SY对大鼠急性脑缺血后24小时炎性损伤有显著的保护作用。
Objective: To explore protective effect of safflower yellow pigment (SY) on inflammatory injury in rats in the different time points after acute cerebral ischemia.Methods: One hundred and eighty healthy male SD rats were randomized divided into 12 groups, including sham groups (3 h, 6 h, 12 h, 24 h) , MCAO groups (3h, 6h, 12h, 24h) , SYgroups (3h, 6h, 12h, 24h) (n=15ineachgroup) .TNF-α, IL-6 and IL-10 levels in rats serum were measured by ELISA .The pathologic morphology in rats brains were observed using HE staining analysis.Results: After being treated with SY, the increased levels of TNF- α (3 h, 24 h) and IL - 6 (3 h, 6 h and 24 h) were significantly inhibted, while IL - 10 (3 h, 12 h and 24 h) were significantly promoted. In addition, the rats brains inflammatory injury in 24th hour after acute cerebral ischemia were obviously relieved.Conclusion: SY can relieve the inflammatory injury in rats brains in the 24th hour after acute cerebral ischemia.
