西黄丸浸提液对人乳腺癌癌前病变细胞mTOR与VEGF表达的影响

目的:探讨西黄丸浸提液对人乳腺癌癌前病变细胞株的影响及其作用机制。方法:以人乳腺癌癌前病变细胞为研究对象,MTT法检测不同剂量西黄丸浸提液对乳腺癌癌前病变细胞活性的影响,并用流式细胞术检测西黄丸浸提液引起的细胞周期及凋亡率,用Western blot方法检测mTOR与VEGF蛋白表达水平,初步探讨其作用机制。结果:与空白对照组比较,西黄丸浸提液10%、15%、20%浓度组及他莫昔芬组的细胞生长OD值减少,G0/G1期细胞数量比例增高,S期、G2/M期细胞数量比例减少,细胞凋亡率增加,mTOR与VEGF蛋白表达水平降低(P〈0.01或P〈0.05)。结论:西黄丸浸提液可抑制乳腺癌癌前病变细胞活力、诱导细胞凋亡,其作用机制可能与其抑制信号分子mTOR与VEGF蛋白的表达相关。

英文摘要:

Objective : To investigate the effects of liquid extract of Xihuang pills on precancerous cells of human breast cancer and its mechanism of action. Methods : The precancerous cells of human breast cancer were divided into control group,Xihuang pill groups, and tamoxifen group. The control group and the tamoxifen group were given normal saline and tamoxifen, respectively, while the Xihuang pill groups were given different doses (10% , 15 % , and 20% ) of liquid extract of Xihuang pills. After treatment, the cell viability was measured by MTF assay. Cell cycle distribution and cell apoptosis were analyzed by flow cytometry. The protein expression of mammalian target of rapamyein(mTOR) and vascular endothelial growth factor (VEGF)was measured by Western blot. Results:Compared with the control group, the Xihuang pill groups and the tamoxifen group had significantly lower optical density, proportion of cells in S phase and G2/M phase, and mTOR and VEGFexpression, and significantly higher proportion of cells in G0/G1 phase and apoptosis rate(P 〈0.01 or P 〈0. 05). Conclusion:Liquid extract of Xihuang pills can inhibit cell viability and induce cell apoptosis in precancerous cells of human breast cancer. The mechanism of action may be related to its inhibitory effect on roTOR and VEGFexpression.
