目的研究二苯乙烯苷(TSG)对H2O2诱导损伤的人脐静脉内皮细胞P选择素和E选择素表达的影响。方法运用逆转录聚合酶链反应(RT-PCR)和酶联免疫吸附试验(ELISA)分别检测P选择素和E选择素的mRNA和蛋白表达。结果 200μmol/L的H2O2作用人脐静脉内皮细胞24 h,P选择素和E选择素的mRNA和蛋白表达水平均明显上调,与空白对照组比较,差异具有显著性(P〈0.01),但TSG预处理人脐静脉内皮细胞4 h后再加200μmol/L H2O2作用24 h,H2O2诱导的内皮细胞P选择素和E选择素的mRNA和蛋白表达水平降低,与H2O2处理组比较,差异有显著性(P〈0.05)。结论 TSG通过降低黏附分子P选择素和E选择素的表达可保护内皮细胞免受氧化应激损伤,影响动脉粥样硬化进程。
Aim To study the effect of 2,3 ,5 ,4'-tetrahydroxystilbene-2-O-β-D-glucoside (TSG) on expression of adhesion molecules P-selectin, E-selectin induced by H2O2 in human umbilical vein endothelial cells. Methods The expression of P-selectin, E-selectin mRNA and protein were detected by reverse transcription polymerase chain reaction (RT-PCR) and enzyme-linked immunosorbent assay (ELISA) respectively. Results Treating endothelial cells with 200 μmol/L H2O2, the expression of E-selectin, P-selectin mRNA and protein level were significantly higher (P 〈 0. 01 ). Pretreated with TSG for 4 h, the expression of P-selectin, E-selectin mRNA and protein induced by H2O2 were significantly inhibited in endothelial cells (P 〈 0.05 ). Conclusions TSG probably can protect human umbilical vein endo thelial cells from oxidative stress by decreasing expression of adhesion molecules.
