生命材料冷冻复温后低温保护剂的去除是影响其低温保存效果的一个很重要的因素.透析方法可以快速有效地去除低温保护剂.为了得到优化的透析清洗程序,变化清洗液以及血液流速,实验测定了透析法清除红细胞低温保护剂甘油过程中细胞渗透压的变化.结果发现:初始时刻透析液侧预填充高渗保护剂溶液以及降低血液和清洗液的流速能有效避免初始时刻渗透压突变,降低渗透损伤;在清洗的后期加快血液和清洗液的流速,可以有效提高清洗效率,缩短保护剂的清洗时间.
The removal of cryopretective agent(CPA) from frozen-thawed biomaterials is one of the main factors that affect the success of a cryopreservation. The dialysis method can remove CPAs quickly and effectively. In order to get the optimized washing procedure, the change in osmolality was measured experimentally with different blood side and dialysate side flow rates and under the conditions of prepriming. The results suggest that. during the initial period of the removal process diminishing the blood side and dialysate side flow rates and prepriming hyperosmotic solution at the dialysate side can effectively avoid the sudden change of the osmolality which can reduce the osmotic injury to the cells; during the later period of the removal process increasing the blood side and dialysate side flow rates can dramatically improve the removal efficiency and reduce the washing time.
