目的观察和分析骨髓干细胞(BMSCs)移植对大鼠急性缺血性肾损伤后肾小管上皮细胞坏死、变性及增殖的影响。方法 Percoll密度梯度离心法分离获取BMSCs。制作大鼠急性缺血性肾损伤模型,通过下腔静脉进行BMSCs移植。分别于缺血再灌注后24h、48h、7d、14d获取肾脏标本,HE染色作肾组织学观察,免疫组织化学染色检测增殖细胞核抗原(PCNA)的表达。结果缺血再灌注后24h、48h,BMSCs移植组肾小管上皮细胞未见坏死及明显变性,而对照组细胞坏死及变性明显;缺血再灌注后7d、14d,BMSCs移植组和对照组肾小管上皮均无细胞坏死,但对照组可见部分细胞胞浆肿胀及间质内少许红细胞;BMSCs移植组较对照组肾小管上皮PCNA阳性细胞数增多,组间差异具统计学意义。结论 BMSCs移植可减少急性缺血性肾损伤的肾小管上皮细胞变性、坏死,而促进肾小管上皮细胞的增殖。

英文摘要:

Objective To investigate the effect of transplanted bone marrow stem cells(BMSCs)on renal tubular epithelial cells after acute renal ischemic injury in SD rats.Methods BMSCs were isolated from the fe-murs and tibiae of male SD rats with Percoll density gradient centrifugation.All model rats were subjected to clamping bilateral renal pedicles for 45 minutes,followed by injection of BMSCs or saline into the inferior vena cava,and were sacrificed to collect the kidneys at 1,2,7,14 days after operation,respectively.All kidney sam-ples were examined by HE staining under light microscope.Immunohistochemical staining was used to examine the express of proliferating cell nuclear antigen(PCNA).Results No necrosis and significant tubular cell degen-eration were observed in kidneys of trasplantation group at 1,2 days after operation.Cell swelling and occasional red blood cell in the interstitium were visible in control group as compared with trasplantation group at 7,14 days.The number of PCNA-positive cells in kidney samples of trasplantation group was more than that of control group at 7,14 days.Conclusions Transplanted BMSCs can attenuate necrosis,degeneration of the renal tubular epithelial cells and accelerate the cell proliferation after acute renal ischemic injury.
