目的探讨miRNA-93在糖尿病肾病患者血清的表达情况及其临床意义。方法用实时荧光定量PCR技术分别检测20例糖尿病肾病患者血清和健康对照组血清中miRNA-93的表达变化。分析其与临床病理参数的关系;运用TargetScan等靶基因预测分析软件预测miRNA-93靶基因。结果与健康对照组相比,糖尿病肾病患者血清中miRNA-93低表达(P〈0.05);尿毒症组患者血清中miRNA-93较非尿毒症组低表达(P〈0.05);生物信息学分析显示为糖尿病肾病中miRNA-93的靶基因为VEGFA、MTHFR、ADRA2B、SDC2、APOB、PPARG。结论 miRNA-93在糖尿病肾病患者血清中低表达,可能通过调控其下游靶基因在糖尿病肾病发生过程中起重要作用。
Objective To study serum microRNA-93 (miRNA-93) expression in diabetic nephropathy pa- tients and its clinical significance. Methods The expression levels of serum miRNA-93 in diabetic nephropa, thy patients and healthy controls were measured by quantitative real-time polymerase chain reaction (qRT- PCR). The relationship between miRNA-93 and the clinicopathological parameters in diabetic nephropathy was investigated. Target genes of miRNA-93 were predicted with bioinformatics softwares. Results Compared to healthy controls, serum miRNA-93 was significantly decreased in diabetic nephropathy patients (P〈0.05). In addition, serum miRNA-93 was also significantly decreased in uremic patients compared with non-uremic pa- tients(P〈0.05). The potential target genes of miRNA-93 identified by bioinformatics softwares include VEGFA, MTHFR, ADRA2B, SDC2, APOB and PPARG. Conclusion Serum miRNA-93 is low- expressed in diabetic nephropathy patients. It may play an important role in the development and progression of diabetic nephropathy through its target genes.
