目的观察比较miRNA-15a和miRNA-17在多囊肾患者家系中的表达水平,探讨miRNA与多囊肾病发生发展的可能关系。方法采用实时定量PCR技术检测多囊肾患者及其家系成员与正常人血清中miRNA-15a和miRNA-17的表达水平。结果所检测的多囊肾患者家系中已出现肾功能损害的多囊肾患者血清miRNA-15a的表达水平明显高于正常人及肾功能正常的多囊肾患者;而miRNA-17在多囊肾患者家系未表达。结论多囊肾患者血清miRNA-15a表达水平与正常人不同,提示miRNA在多囊肾的发生发展过程中可能起重要调控作用,值得深入研究。
Objective To explore the relationship between serum miRNA and the occurrence or development of polycystic kidney disease in patient families.Method Serum levels of miRNA-15a and miRNA-17 of patients with polycystic kidney disease including their family members and the controls were measured by real-time PCR.Results Compared with the controls,the cases of polycystic kidney disease with renal dysfunction showed a higher level expression of miRNA-15a,but miRNA-17 expression were not found in the patient group.Conclusions Serum miRNA-15a level in the patient with polycystic kidney disease is different from healthy control.These results imply that miRNA may play important regulatory role in the occurrence and development of polycystic kidney disease.
