目的:研究阿司匹林对心肌梗死后诱导心肌纤维化的防治作用。方法:采用冠脉结扎复制大鼠急性心肌梗死模型,随机分为模型组(Mod)、阿司匹林组(30mg·kg^-1)及假手术组(Sham)。分析模型复制4周与8周后,大鼠血流动力学(左心室舒张末期压、左心室收缩压、左心室平均压、左心室最大收缩速率及左心室最大舒张速率)、心肌重构指数(左心室前壁、左心室直径及心脏长轴)的变化,并进行大体解剖学观察及病理组织学检查。结果:模型复制4周与8周后,血流动力学指标紊乱、心室收缩与舒张功能障碍、心肌重构指数增加。大体解剖学及病理组织学检查提示形成心肌纤维化;给予阿司匹林后对上述指标具有显著的改善作用(P〈0.05),能改善心功能,减轻心肌纤维化及心室重构。结论:阿司匹林对实验性心肌纤维化具有一定的防治作用。
OBJECTIVE To investigate the protective effect of aspirin on experimental myocardial fibrosis induced by acute cardiac infarction through the ligation of coronary artery in rats. METHODS The rats were randomly allotted to 3 groups as follows: sham operated group,model group and the aspirin group (the coronary artery was ligatied + 30 mg·kg^-1 aspirin). The animals were lived four or eight weeks after operation. The cardiac infarction was reproduced by ligating rat coronary artery.Four and eight weeks after the ligation of the coronary in rats, the following indexes were detected, hemodynamics including(left ventricular end-diastolic pressure,left ventricular systolic pressure,left ventricular mean pressure, maximum rate of left ventricular contraction, and left ventricular maximum rate of diastolic), cardiac remodeling indexes including (anterior left ventricular, left ventricular internal diameter, heart-long axis). And then the gross anatomy observation and histopathologic changes were recorded. RESULTS After four and eight weeks,the myocardial fibrosis was reproduced according to gross anatomy, and his- topathologic examination. Ventrieular systolic and diastolic dysfunctions and increasing of reconstruction indexes were observed as well. There were significant ameliorations of the indexes after the rats were treated with aspirin. CONCLUSION Aspirin can ameliorate experimental myocardial fibrosis induced by acute cardiac infarction.
