目的研究溶血磷脂酸(lysophosphatidic acid,LPA)受体在人卵巢癌组织中基因的表达及意义,探讨LPA对人卵巢癌组织中Bcl-2和Bax基因表达的影响及意义。方法采用反转录聚合酶链反应法检测90例卵巢组织(包括正常卵巢组织、卵巢良性肿瘤组织、卵巢上皮癌组织)中LPA1、LPA2和LPA3基因的表达,分析LPA1、LPA2和LPA3基因表达与Bcl-2、Bax基因表达的相关性。结果在卵巢癌组织中LPA2和LPA3基因表达明显高于卵巢良性肿瘤和正常卵巢组织(P〈0.05)。卵巢良性肿瘤和正常卵巢组织中LPA1基因表达明显高于卵巢癌组织(P〈0.05)。LPA2和LPA3基因表达与Bcl-2基因表达呈显著正相关(r=0.308,P〈0.01),与Bax基因表达呈显著负相关(r=-0.369,P〈0.01)。结论LPA及其受体可能在卵巢癌的发生发展中起重要作用,LPA促进卵巢癌发展的机制之一可能是通过LPA2和LPA3影响Bcl-2和Bax表达,从而改变Bcl-2/Bax比例,抑制卵巢癌细胞的凋亡,促进卵巢癌的发展。
Objective To investigate: the expressions of lysophosphatidic acid (LPA) receptors at mRNA levels and their biological significance in different human ovarian epithelial ovarian neoplasms,and to study the relationship between the expressions of LPA1, LPA2, LPA3 genes and Bcl-2,Bax genes. Methods Reverse transcription-polymerase chain reaction(RT-PCR) was used to measure the expression levels of LPA1, LPA2, LPA3 and Bcl-2,Bax mRNA in normal ovary,ovarian benign tumor and ovarian cancer. Results LPA2 ,LPA3 and Bcl-2 expression ratios significantly increased in cancer, compared to those in non-cancerous state ( P 〈 0.05). Bax expression ratios significantly decreased in cancer compared to those in non-cancerous state( P 〈 0.05). A significant positive correlation in the expression ratios between LPA2 or LPA3 and Bcl- 2,and a significant negative correlation between LPA2 or LPA3 and Bax was found( r =0. 308, P 〈0.01; r =-0. 369., P 〈0.05) in patients with cancer. Conclusion LPA and its receptors may play an important role in the ovarian cancer; LPA2 and LPA3 maybe involved in Bcl-2 and Bax expression mediated by LPA signals in human ovarian oncogenesis. LPA changed the ratios of Bcl- 2/Bax though LPA receptors,which inhibited the apoptosis of ovarican cell and promoted ovarian cancer progression.
