目的 探讨磷脂酰肌醇3-激酶/丝氨酸-苏氨酸蛋白激酶(P13K/Akt)信号转导通路阻断剂LY294002对卵巢癌细胞顺铂(DDP)敏感性的影响。方法DDP和LY294002单独或联合作用人卵巢癌细胞,MTT法检测细胞生长抑制率,倒置显微镜观察凋亡细胞形态,流式细胞术检测凋亡细胞。结果加用LY294002后,DDP对卵巢癌细胞的抑制作用增强,细胞凋亡率上升。结论PI3K/AKt信号转导通路阻断剂可有效提高卵巢癌细胞对DDP的敏感性,在卵巢癌治疗中与DDP有一定协同作用。
Objective This study aims to investigate whether inhibition of PDK/Akt signal pathway increases the effect of DDP on ovarian cancer cells. Methods The inhibition effects of DDP alone or in combination with PI3K/Akt inhibitor LY294002 on proliferation of 3AO and SKOV3 cells were measured by MTr assay;Optic microscope was used to detect the morphological changes of 3AO and SKOV3 cells. Effects of DDP alone or in combination with LY294002 on apoptosis of 3AO and SKOV3 ceils were detected by flow cytometry. Results The inhibition ratio of proliferation of 3AO and SK- OV3 ceils was increased signifieknfly by DDP in combination with LY294002 ;The apoptotie ration of 3AO and SKOV3 cells was increased by DDP in combination with LY294002. Conclusions LY294002 can significantly enhance sensitivity to the DDP. Inhibition of PI3K signal transduction pathway could have a significant effect on treatment of ovarian carcinoma
